Use this iRule:

when HTTP_REQUEST {

    Check if requested host equals to testing.com
   if {[string tolower [HTTP::host]] eq "testing.com"}{

       Replace the host header value with testing.com:5060
      HTTP::header replace Host "testing.com:5060
   }
}
